The XZ Dialysis System
Dialysis protein crystallization is a well-established method for protein crystallization developed during the early days of structural biology. dialysis is the only technique that allows crystallization of a biological macromolecule by either salting-in or salting-out. The disposable XZ microfluidic dialysis technology,has been developed to specifically address these issues. With the XZ system, dialysis protein crystallization trials as low as 6 nl/reaction can be easily and quickly setup.

The XZ dialysis protein crystallography plate is composed of a microfluidic plate bonded to an open bottom microtiter plate. The microfluidic plate contains a microfluidic network on the bottom which delivers a protein sample solution to the dialysis chambers. The microfluidic plate is sealed on the bottom with a clear film and on the top with dialysis membrane discs over the dialysis chambers. The dialysis disc side of the microfluidic plate is then bonded to an open-bottom microtiter plate.
Crystallization solution is first dispensed into the upper reservoirs of the X plate. The plate is then connected to a vacuum pump and the system evacuated. While under vacuum, the protein is introduced into the microfluidic circuit where it is carried throughout the plate, filling each of the dialysis chambers. A subsequent, immiscible purge solution is then applied to the microfluidic circuit to seal the chambers from one another. The process can be easily completed in 10 minutes per plate.
We highly recommend the use of the Salt-in/Salt-out crystallization screen with the XZ system.
The XZ Dialysis plates are available in four formats, summarized in the table below. Plates differ in the volume per reaction chamber and in the number of reaction chambers per plate. The XZ kit comprises 3 each of the 96-well formats to permit first time users to evaluate which screening volumes are most appropriate for their needs.
